1.  A substrate “A” can be converted to a product “B” by Enzyme 1.  Substrate “A” can also be converted to “C” by Enzyme 2.  

The Kmof Enzyme 1 = 23 mM

The Km of Enzyme 2 = 76 mM

The Vmax of Enzyme 1 = 133 μmoles of product/minute

The Vmaxof Enzyme 2 = 133 μmoles of product/minute

(a) Draw a rough enzyme kinetics [S] vs. v0graph with the above data.  Show the curve for Enzyme 1 first and then repeat for Enzyme 2.

(b) At 23 mM substrate concentration [A], what is the velocity of Enzyme 1?

(c) At 76 mM substrate concentration [A], what is the velocity of Enzyme 2?

(d) At 23 mM substrate concentration [A], what is the velocity of Enzyme 2?

(e) You do an experiment to test these enzymes.    In test-tube #1, you have enzyme 1 and provide it with 23 mM of substrate A.  In test-tube #2, you have enzyme 2, and you provide it with 23 mM of substrate A.  

         (i) How many μmoles of “B” are made in test-tube #1 after running the reaction for 1 minute?

         

         

         (ii) how many μmoles of “C” are made in test-tube 2 after running the reaction for 1 minute?

2.  A substrate “A” can be converted to a product “B” by Enzyme 1.  Substrate “A” can also be converted to “C” by Enzyme 2.  

The Kmof Enzyme 1 = 15 mM

The Km of Enzyme 2 = 34 mM

The Vmax of Enzyme 1 = 180 μmoles of product/minute

The Vmaxof Enzyme 2 = 180 μmoles of product/minute

(b) You do an experiment to test these enzymes.    In test-tube #1, you have enzyme 1 and provide it with 15 mM of substrate A.  In test-tube #2, you have enzyme 2, and you provide it with 15 mM of substrate A.  

         (i) How many μmoles of “B” are made in test-tube #1 after running the reaction for 8 minutes?

         

         

         (ii) How many μmoles of “C” are made in test-tube 2 after running the reaction for 8minutes?
