This summer you interned in a lab that works with mice. You notice that a mutation has arisen in your mouse colony that leads to significantly increased susceptibility to infections. Most of these mice die early in life due to infections. You perform a quick assay of the blood and find there are no antibodies present.

a). (5 points) Describe briefly how you could show whether all leukocytes are absent, only lymphocytes, only T cells, or only B cells?

b). (10 points) Describe an experiment you could use to identify if the defect is in these mice is in their bone marrow’s hematopoietic cells OR non-hematopoietic cells (e.g. stromal or reticular cells). Be sure to include appropriate controls and a description of what you would expect to see for each possibility (in other words, what would you expect to see if the defect was in HSCs? What would you expect to see if the defect is in other non-hematopoietic bone marrow cells?
